Microspectrophotometric DNA analysis in ulcerative colitis. (Report 1). With special reference to the assessment of optimum section thickness.

A study was conducted to assess the optimum thickness of paraffin-embedded sections to determine their true DNA content using the cytophotometric technique in 35 colonic samples from 23 patients with ulcerative colitis and 10 non-colitic patients with colorectal cancer. Sections 4, 7 and 10 μm thick were cut from each sample. The cellular DNA values of these sections were compared with smears urom cells detached from the same paraffin-embedded tissues. The modal DNA values for the four methods were similar. In contrast, the frequencies of cells with a DNA content higher than 6 c (polyploid cells) did not coincide. The frequencies of false-negative cases which had polyploid cells in smears but not in sections were 8.6, 0 and 5.7 in the 4, 7 and 10 μm sections, respectively. The frequencies of false-positive cases were 14.3, 20 and 25.7, but 6.3, 6.7 and 18.8 in the inactive specimens. Therefore 7 μm sections showed the highest sensitivity and specificity. We suggest that the microspectrophotometric DNA content of ulcerative colitis specimens is better analyzed using 7 μm sections.
