• Twenty rats aged 6 weeks were treated with weekly subcutaneous injeceion of 15 mg/kg of DMH for 30 weeks. Five rats were treated with injection of saline and served as controls.<BR>One-hundred-and-three tumors developed in the colon of DMH- treated animals, whereas no tumor occurred in the colon of controls. Eighty-four out of 103 tumors developed in the colon of DMH-treated animals were invasive carcinoma and most of them were highly differentiated adenocarcinoma. These tumors occurred mostly in the proximal and mid-colon.<BR>A typism of epithelial cells and/or crypts was found in 85 out of 100 specimens taken from the flat mucosa of the colon of DM-treated animals, whereas no atypism was found in 25 specimens taken from the flat muosa of the controls.<BR>Analysis of DNA histogram disclosed that there was a correlation between the degree of the atypism and appearance of cells with polyploidy. Appearance of cells with polyploidy in the upper and middle portion of the crypt was characteristic of flat mucosa of the colon of DMH-treated animals.
