Neurosecretion and Anterior Pituitary Cells:III. Correlation between neurosecretion and cytological changes in the anterior hypophysis following castration in rabbits

In castrated rabbits, neurosecretion is gradually augmented in the first month after castration. Great amounts of neurosecretory granules are released from the hypothalamic-neurohypophysial neurosecretory system (HNS) into the blood vessels and into the adeno-hypophysis. During later post-operative days, hypothalamic neurosecretory cells underwent, by turn, two phases, viz., a long hypersecretory and a short hyposecretory phase (or a resting period), suggesting a cyclic fluctuation of neurosecretory activity in castrated rabbits. At 8 months after castration, regressive degeneration occured in cells of both supraoptic and paraventricular nuclei and neurosecretory granules decreased greatly in the HNS. These changes in the neurosecretory system after castration were followed by histological changes in the adenohypophysis. Gonadotroph cells (positive in PAS-staining and negative in AF-staining) increased gradually in number and size in post-operative days and the secretion of granules was augmented em. Thyrotroph cells reduced in size, number, and density of granulation in post-operative days. In the 8th post-operative month when a great decrease was shown in neurosecretory granules in the HNS, these cells increased greatly in size and number. After castration the acidophils of the adenohypophysis increased slightly in number, while neuro-secretory granules increased in the first several days. Then the acidophils decreased in number by the increase in number and size of gonadotroph cells in later post-operative days.From the results obtained, it was postulated that the pituitary-gonadal system was under the control of the hypothalamic neurosecretion, and that the increase of neurosecretory substance in the adenohypophysis stimulated the production and release of the gonadotrophic hormones of the anterior pituitary.
