In their previous paper, Koyanagi et al. reported the results of feeding experiments carried out on chickens fed fish oil diets. It was the purpose of these experiments to examine whether or not the so-called anti-fatty liver factors might prevent the deposition of a fatty substance in the liver.In this connection, the present authors conducted further experiments successively by using laying hens.Pullets were fed a standard breeder ration for approximately 9 weeks after hatching and divided into 5 groups of 19 each. One group was fed a basal diet as control; the other 4 groups received similar diets containing 10 per cent sardine oil. Besides, in 3 of the 4 experimental groups, the diet was supplemented with choline, choline plus methionine, and choline plus fish meal, respectively, as lipotropic factors.After 6 months observation, 10 birds were killed in each group for postmortem examination.The results obtained are as follows.1) During the experimental course, pullets of the groups fed fish oil diets died in succession from hemorrhage in the liver. Necropsy revealed discoloration and, to a certain extent, swelling in all livers and some kidneys. Besides, metamorphotic, abnormal ovarian follicles were observed in many cases.2) Marked and rather characteristic changes of the liver as mentioned below were confirmed histologically in all the groups, except the control.a) Local lesions of various sizes were formed. They consisted largely of cells resembling the liver cells around the lesion. Their cytoplasm was filled with dark, blue or grayish granules after H. E. staining. These granules were stained red by the acid-fast bacteria staining technic and Mallorys hemofuscin method. The parenchyma was usually dotted with these lesions which appeared in an isletlike arrangement.b) Necrotic foci were present in various parts, Diffuse necrobiotic or necrotic figures were also noted in some cases.c) Marked deposition of acid-fast, ceroid-like pigment granules was noted in the liver cells and macrophages especially in perivascular regions.3) The supplement of any lipotropic factor to the diet did not bring about the expected results, i. e., prevention of any changes in the liver and other viscera.
