The role of estrogen in the formation of the chicken's medullary bone was histologically investigated via the administration of an antiestrogen (tamoxifen citrate). The antiestrogen was given to capons in which medullary bone formation had been induced in the marrow cavities via injections of both estrogen and androgen. Antiestrogen was also given to laying hens with an egg in the magnum or uterus (shell gland) of the oviduct. The medullary bone developed rapidly in capons which were treated with estrogen and androgen, whereas administrations of antiestrogen to these capons prevented osteoblastic formation and induced osteoclastic resorption, resulting in a rapid reduction of the matrix of the medullary bone. In laying hens, medullary bone formation was accompanied by the appearance of numerous osteoblasts which occurred when an egg was in the magnum of the oviduct, whereas administrations of antiestrogen during this period resulted in a significant increase in the number of osteoclasts. Despite the marked appearance of osteoclasts in the medullary bone surface, antiestrogen administrations caused further increases in the number of osteoclasts, when an egg was in the uterus of the oviduct. These findings indicate that osteoclastic resorption of the medullary bone occurred in laying hens which were administered with antiestrogen. From the above results, it may be concluded that when the action of estrogen is blocked by antiestrogen administrations, osteoblastic formation is essentially inhibited, but osteoclastic resorption is strongly precipitated in the medullary bone. However, administrations of antiestrogen when an egg is in the magnum or uterus of the oviduct caused a marked reduction in the volume of plasma calcium despite the induction of osteoclastic resorption of the medullary bone.
