To investigate the relation between disappearance of dry matter (DM) and acetyl bromide lignin (ABL) of tropical grass, an experiment measuring the disappearance of DM and ABL of Rhodes grass (Chloris gayana) was conducted using in vitro incubation with rumen fluid-pepsin or pepsin-cellulase respectively. Rumen fluid was collected from three goats fitted with rumen fistulae and fed a hay. Onozuka cellulase SS (P-1500) was used in pepsin-cellulase method. ABL was determined by MORRISON'S method. Materials were leaf and stem of an internode elongation (immature) and of a mature stage of regrowth. The results obtained were as follows: In leaf and stem, ABL contents were higher than acid detergent lignin (ADL) contents, and less value of ABL/ADL in mature grass comparing with immature grass suggested that more amount of lignin-like substance of immature grass were dissolved in acid detergent solution than those of mature grass. In incubation of leaf and stem of immature and mature materials by rumen fluid-pepsin method, DM disappearance of them were 70.2%, 63.7%, 55.4% and 37.8% respectively, and ABL disappearance of the same materials were 46.5%, 34.6%, 31.49/6 and 19.1%. By pepsin-cellulase method, DM disappearance of the materials were 62.0%, 50.6%, 46.2% and 27.5% respectively, and there occurred a disappearance of ABL that might be induced by morphological uncertainty of lignin in the plant tissue and/or broad spectrum action of Onozuka cellulase SS (P-1500). In this case, though the values of ABL disappearance were lower than the values by rumen fluid, they were 30.3%, 16.4%, 15.7% and 6.1%. Comparison of disappearance of DM and ABL by rumen fluid-pepsin method with those by pepsin-cellulase method suggested that disappearance of ABL affected the digestion of lignified cell wall substance particularly in mature grass. There was positive correlation (P<0.05) between disappearance of DM and ABL in incubation by rumen fluid-pepsin method.
