Studies on the Agglutinability and Hemolytic Activity of Various Phytohemagglutinins to the Red Blood Cells of Mice

Many mutants, inbred and congenic strains of mice have been utilized as excellent experimental animals for the investigations on immunology or immunogenetics. An H-2 locus in mice is the major histocompatibility complex (MHC) system and the antigens controlled by H-2 locus are on the red blood cells of mice. Blood group systems other than H-2 locus have not been studied in detail. It is known that extracts of certain plant seeds agglutinate the red blood cells of various animals, including human beings. The effective factors are known as phytohemagglutinins or lectins, and some of them have practically been used for blood grouping. In the present studies, extracts prepared from 111 different plants including 39 species of Leguminosae were tested for their agglutinability against BALB/c-hr strain, dd strain maintained in closed colony and wild mice. Based on the results obtained, these extracts could be classified into following six groups: Group I., 24 extracts which did not agglutinate and not hemolyze the red blood cells of all mice tested; Group II, 63 extracts which agglutinated and did not hemolyze the red blood cells of all mice tested: Group III, 5 extracts which did not agglutinate and hemolyzed the red blood cells of all mice tested; Group IV, 1 extract which agglutinated and hemolyzed the red blood cells all mice tested; Group V, 17 extracts which agglutinated the red blood cells of some mice, but did not agglutinate the red blood cells of the remainders and did not hemolyzed the red blood cells of all mice tested; Group VI, 1 extract which agglutinated the red blood cells of some mice, but did not agglutinate the blood cells of the remainders and hemolyzed the red blood cells of some mice, but did not hemolyze the red blood cells of the remainders. Some properties of the agglutinogens reacting with Helianthus annuns PHA and Allium sativum PHA and of the red blood cells hemolyzed by the extract from Allium sativum were also studied using many mice including BALB/c-hr strain, dd strain and wild mice. It was suggested that the disappearance of the agglutinogen reacting with Helianthus annuns PHA in females of dd strain was in relation to the pregnancy. The presence of the agglutinogen reacting with Allium sativum PHA was genetical and the agglutinogen was in 5-9 weeks old mice but after 10 weeks old the agglutinogen disappeared in all mice tested. The properties of the red blood cells hemolyzed by the extract from Allium sativum were assumed to be genetical, but the mode of inheritance was not revealed.
