This experiment was carried out to examine the effect of methionine sulfoximine (MSM), glutamine synthetase inhibitor, on glutamine and uric acid synthesis in the chicken fed a 5% protein diet. Plasma glutamine concentration decreased significantly and blood ammonia concentration increased significantly within 3hr after a single intravenous injection of 100mg of MSM/kg BW, whereas plasma uric acid concentration was not affected significantly by the treatment. When 5% glucose was infused, the pretreatment of MSM caused significant decreases in liver glutamine synthetase activity and plama glutamine concentration, and no significant decreases in glutamine content of liver and in plasma and urinary uric acid levels. On the other hand, the pretreatment of MSM significantly increased ammonia concentration in the blood, liver and urine. The infusion of ammonium acetate into the chicken pretreated with MSM slightly increased uric acid concentration in the blood plasma and urine without causing significant changes in glutamine concentration in the blood plasma and liver. Glutamine infusion in the MSM-treated chickens markedly increased urinary uric acid excretion and plasma uric acid concentration, and also remarkably increased ammonia concentration in the liver and urine. These results suggest that the MSM treatment in the chicken fed a low protein diet results in an almost complete inhibition of glutamine synthesis and in no significant decrease in uric acid synthesis, and that uric acid synthesis in such chickens is increased slightly by ammonia infusion and greatly by glutamine infusion.
