The present experiments were carried out to clarify the relationship between the nucleation temperature of diluted bull semen and the cooling rate around O°C, and to clarify the effects of the nucleation temperature on motility of bull spermatozoa after frozen to -196°C and thawed. Bull semen was diluted with egg yolk-sodium citrate dilutor to contain 7% (v/v) glycerol and cooled with liquid nitrogen vapour. Temperature changes of diluted semen were measured with a thermoelectric thermometer fitted with an iron-constantan thermocouple. The results were as follows. 1. When diluted semen sealed in the vinyl straw (1ml) was cooled at the rates of 0.5-109.8°C/min around 0°C, the nucleation temperatures ranged between -4.0°C and 13.8°C. After diluted semen sealed in the glass ampule (1ml) was cooled at the rates of 0.6-21°C/min around 0°C, the nucleation temperatures ranged between -3.0°C and -9.2°C. The nucleation temperatures of diluted semen were generally lower at slow cooling rate around 0°C than at rapid cooling rate under the present experimental conditions, though the supercooled state of semen was relatively unstable. 2. Similar motility of frozen-thawed spermatozoa was obtained when they were cooled at the similar cooling rate after the break of the supercooled state even if the nucleation temperature of semen varied. 3. There was no difference in motility of frozen-thawed spermatozoa whether supercooling of semen was broken spontaneously or with ice-seeding.
