Two phosphonosphingolipids,ceramide aminoethylphosphonates named CAEPn-1 and CAEPn-2 were purified from the pearl oyster,Pinctada martensii by successive column chromatography on ion exchange Sephadex (QAE-and DEAE-Sephadex) and silicic acid (Iatrobeads) and their aliphatic components were determined by converting to various derivatives.The numbers of carbons and hydroxylated positions of the fatty acids were confirmed by gas chromatograph-mass spectrometry as their methyl ester derivatives.On the other hand, the numbers of carbons and double bonds of the sphingoids were confirmed by gas chromatograph-mass spectrometry as their trimethylsilyl ether derivatives.Double bond positions in the aliphatic chains were also confirmed by gas chromatographic and gas chromatograph-mass spectrometric identifications of the fatty acid products after periodate-permanganate oxidation of the ceramides derived from the CAEPn by hydrogen fluoride degradation.The major fatty acids of CAEPn-1 were hexadecanoic (82.0%)and heptadecanoic acid (13.9%), and those of CAEPn-2,2-hydroxyhexadecanoic (84.1%) and 2-hydroxy heptadecanoic acid (15.9%).The major sphingoids of CAEPn-1 were octadeca sphinga-4-enine (24.7%),4,8-dienine(40.1%) and 4,8,10-trienine (24.6%),and those of CAEPn-2,octadeca sphinga-4,8,10-trienine (29.2%)and its 9-methyl homologue (58.6%).
