Muscle strength exercises have not adequately accounted for the effects associated with the differences in ages of the participants or the objective muscles. We investigated the effects of endurance training on young and old rat hindlimb muscles. Young (10-week-old) and old (104-week-old) male Wistar rats were randomly assigned to either a control group or exercise group [young control (YC), old control (OC), young exercise (YE), old exercise (OE)]. Exercise training consisted of 4 weeks of treadmill running (20 m/min, ±0% grade, 30 min/day, 6 days/week). Thereafter, the soleus (SOL) and extensor digitorum longus (EDL) were measured for their in vitro isometric contractile properties, cross sectional area (CSA) of muscle fibers, and myosin heavy chain (MHC) isoform composition. The specific twitch tension of the SOL was significantly increased in the YE to the YC, although the SOL and EDL were significantly decreased in the OE to the OC. The CSA of the SOL tended to increase in the exercise groups of both ages, but that of the EDL tended to decrease. There were no significant differences in the relative composition of MHC, although both the SOL and EDL tended to make the transition to slow type myosin from fast type myosin in the exercise groups of both ages. These findings demonstrate that in the SOL of young rats muscular hypertrophy and muscle strength are induced by endurance training, but in old rats, both the SOL and EDL were damaged by this training and they showed decreases in function.
