[原著]Estimation of Sodium- Potassium ATPase Activity of Skeletal Muscle Hemogenate and Washed Particle

Applicability of ouabain and vanadium for estimation of Na-K ATPase of crude fractions of skeletal red and white muscles was examined. More than 3 mM ouabain or 300 nM vanadium significantly reduced magnesium-activated ATPase activity of skeletal muscle homogenate and washed particle. Na-K ATPase activity of red muscle homogenate estimated using 1 mM ouabain was about 2-fold higher than that of the washed particle, while the Na-K ATPase activity of white muscle homogenate was about half in comparison with that of the washed particle. This indicates differences in changes of Na-K ATPase activity between skeletal red and white muscles during preparation of washed particle from the homogenate. Na-K ATPase activity of skeletal muscle homogenate estimated using 100 nM vanadium showed a similar activity to that of the washed particle, which was lower than that estimated by 1 mM ouabain. The present results suggest applicability of ouabain as a tool for estimation of Na-K ATPase in skeletal muscle crude fraction.
