Aldosterone level in plasma was estimated during the course of endotoxin shock of rabbits. The influences of the renin-angiotensin system, hypophyseal ACTH, plasma potassium level and serotonin upon plasma aldosterone level were investigated. The results obtained were as follows ; 1) Plasma aldosterone concentration was increased for 8 hours following endotoxin administration in non-pretreatment rabbits. On the other hand, in the renin blocked rabbits by ligation of renal vessels, plasma aldosterone concentration was increased significantly 30 minutes after endotoxin administration and it was also increased 30 minutes after endotoxin administration in the rabbits in which ACTH was suppressed by pretreatment with dexamethasone after renal vessel ligation for renin blocking. 2) Plasma potassium level was decreased for 60 minutes after endotoxin administration and later was increased up to the level observed before endotoxin administration. No significant change was observed in plasma sodium level. 3) After intravenous injection of serotonin, plasma aldosterone concentration was increased and plasma potassium level was decreased in the renin blocked, ACTH suppressed rabbits. 4) An addition of serotonin to the incubation medium showed an increasing effect on aldosterone output of the adrenal capsular cells in vitro but endotoxin showed no effect in the same experiment. 5) Half-life of exogenous aldosterone became longer during the course of endotoxin shock in the adrenalectomized rabbits. From these results, it was suggested that elevation of plasma aldosterone levels in the earlier stage during the course of endotoxin shock was correlated with the increase of serotonin level in plasma after injection of endotoxin and also with the prolongation of aldosterone, independent of any of the activations of the renin-angiotensin system and hypophyseal-adrenocortical mechanism, and elevation of plasma potassium level.
