Early and late afterpotentials of intact and T-disrupted frog sartorius muscles immersed in NaCl- or sucrose-hypertonic Ringer solution were studied and the following results were obtained. 1) The amplitude of the early afterpotential was enhanced about 7 mV in NaCl-hypertonic Ringer solution and about 17 mV in sucrose-hypertonic Ringer solution. The duration of the early afterpotential showed a tendency of prolongation in both hypertonic Ringer solutions. 2) The action potential in T-disrupted muscle was not followed by a normal early afterpotential, but rather by a small hyperpolarization. 3) The early afterpotentials were not summarized, if the action potentials were evoked by repetitive stimulation. 4) In solutions in which the tonicity was increased with NaCl or sucrose, the amplitude of late afterpotential was increased by increasing the number of stimuli. The extent of enhancement in NaCl-hypertonic Ringer solution was larger than that in sucrose-hypertonic Ringer solution. The half-decay time of the late afterpotential was prolonged by increasing the number of stimuli in both sucrose-hypertonic and NaCl-hypertonic Ringer solutions. The extent of prolongation in the former was larger than that in the latter. 5) Late afterpotential was not observed in T-clisruptecl muscles, although they were exposed to NaCl-hypertonic Ringer solution. On the basis of these results, the mechanism of origin of early and the late afterpotentials was discussed.
