The present report is concerned with the origin and the fate of the oval cells induced by 3'-methyl-4-dimethyl-aminoazobenzene (DAB) and alpha-naphthyl-isothiocvanate (ANIT) in rat liver. The following results were obtained : (1) Within one or two weeks, oval cells appeared in the periportal areas in both DAB and ANIT fed rats. Tubules were formed which were connected with cholangioles and proliferated along the space of Disse between hepatocytes and sinusoidal lining cells. Their ultrastructural characters were similar to those of cholangiolar cells and did not show any remarkable glucose-6-phosphatase (G-6-Pase) activity. (2) In the 4th week, oval cells increased in number in both groups of rats, but oval cells induced by DAB and those by ANIT were quite different. In DAB fed rats, some diversities in morphology of oval cells appeared, some of which showed round nuclei and relatively ample cytoplasm, with an appearance of small hepatocytes. Ultrastructurally, some oval cells were similar to cholangiolar cells while others showed various cytostructural characteristics between cholangiolar cells and hepatocytes. A variation in intensity of G-6-Pase activity, almost proportional to their degree of morphological resemblance to hepatocytes was seen. On the other hand, oval cells induced by ANIT formed relatively large tubules associated with various amounts of connective tissue. Ultrastructurally, these were similar to cholangiolar cells and showed negative or weak G-6-Pase activity of the same degree as seen in normal cholangiolar cells. (3) In the 6th week, in DAB fed rats, small hepatocytes showed an increase which had a marked G-6-Pase activity, corresponding to the decrease of oval cells with cholangiolar features. In ANIT fed rats, some oval cells formed larger tubules with increasingly abundant connective tissue compared with those in the 4th week. However, there was no evidence of transition from oval cells to hepatocytes as seen in DAB fed rats.
