The reactivity of isolated rat parotid cells to α- and β-adrenergic agents was investigated. To avoid diminution of cellular responses to the drugs, isolated parotid cells were carefully prepared by a method which modified the duration of trypsin treatment and the buffer system of the incubation media in the original method of Mangos. Responses of the cells to the drugs were as follows :i) Isoproterenol (ISP) increased amylase release from the cells when applied in concentrations greater than 10-9M, with its maximum effect being at 10-4M. ISP-induced accumulation of cyclic AMP reached the maximum 2 min after administration of the drugs. ii) Methoxamine (10-6-10-4M) increased amylase release from the cells, but the degree of the increase was smaller than that by ISP. The addition of methoxamine into medium did not alter the basal level and ISP-induced elevation of cyclic AMP in the cells. iii) Methoxamine additionally increased ISP-induced amylase release. iv) Methoxamine-induced amylase release was reversed to the level of basal release by removal of calcium from the incubation medium, but ISP-induced enzyme release was unaffected. It was concluded that isolated parotid cells could be employed as a useful tool in the analysis of the action of various adrenergic agents.
