By using anti-synthetic bradykinin (BK) antibody in combination with 125I-tyrosyl-bradykinin, a sensitive and specific radioimmunoassay method for urinary kinins has been developed, and a simplified assay system for the measurement of urinary kallikrein activity by utilizing this kinin radioimmunoassay has also been established. In the radioimmunoassay system of kinin, the dose-response curves of BK, lysyl-BK and methionyllysyl-BK were almost identical. Therefore, it should be thought that the estimated values would exhibit the total kinins in urine. Since a chromatographic study did not demonstrate any substance in urine samples interfering in our assay system, the assay was performed without any extraction procedure. On the other hand, urinary kallikrein activity was measured as the kinins generated by incubation of 50μl of urine samples with 1500ng of kininogen. Since the cross-reactivity of kininogen in our kinin radioimmunoassay system was not recognized at doses up to 1.0μg, the amount of untreated kininogen in the radioimmunoassayed samples did not interfere with the measurement of kinins. This eliminated the necessity for a kininogen extraction procedure. A highly significant correlation was observed between the urinary kallikrein activities determined by our assay system (kininogenase activity) and those by the esterolytic assay. Employing these methods, measurements were taken from normal subjects and patients with various disorders, with the urinary kinins excretion and the urinary kallikrein activity in the normal subjects being 34.5±3.0μg/day and 2.9±0.4μg/min/day, respectively.
