The effects of labetalol on salivation in mice were investigated and following results were obtained. 1) While pilocarpine-induced salivation was not affected by pretreatment with labetalol (1 mg/kg and 10 mg/kg, i. v.), noradrenaline- and adrenaline-induced salivation was inhibited significantly by this pretreatment. Salivary responses to isoproterenol, in doses less than 2.0 mg/kg (i. p.), were inhibited with pretreatment of 1 mg/kg labetalol. However, the responses to isoproterenol in the dose range 2.0 mg/kg to 10.0 mg/kg were potentiated inversely by the pretreatment. 2) Unlike adrenergic agents, labetalol itself caused a weak salivary response after a latent period (about 30 min). This labetalol-induced salivation was markedly inhibited by chlorpromazine and haloperidol, but not affected by the pretreatments with reserpine and 6-hydroxydopamine. From the above results, it was concluded that labetalol has both alpha- and beta-adrenergic blocking actions on mouse salivary glands, and in addition to those actions, it has a weak salivary action which may be caused through activating dopamine receptors.
