In the present animal experiment, the mechanisms for immunosuppresive effect of cyclophosphamicle (CY) and the relationship between a granulomaformation and a subpopulation of lymphocytes were investigated by studying the influence of CY on delayed-type hypersensitivity (DTH) in mice sensitized with the killed mycobacterium tuberculosis intravenously. Results of the above experiment showed that granulomaformation ran almost paralell with organ weight/body weight ratio, with the Migration Inhibition Test with alveolar cells and with the foot pad reaction. These reactions were prominently suppressed by the administration of CY and the suppressive effects were recovered by transfer of of sensitized spleen cells. However, they were not recovered by anti-θ-serum treated sensitized cells. From above results, it was clearly ascertained that these suppressive effects of CY depended upon damage to T-cells, and was suggested that T-cells played an important part in these DTH reactions such as granulomaformation, organ weight/body wight ratio, the macrophage Migration Inhibition Test and foot pad reaction.
