The cDNA fragments for indoleamine dioxygenase-like myoglobins from the abalones Nordotis gigantea and Nordotis discus were amplified by polymerase chain reaction (PCR). The amplified products were subcloned in the SmaI site of pUC18 and the sequences of 1455 and 1247 nucleotides, encoding 373 and 304 amino acid residues, respectively, were determined. The cDNA-derived amino acid sequences of N.gigantea and N.discus myoglobins differ from that of N.madaka myoglobin only in two amino acids [Suzuki, T. (1994) J.Protein Chem. 14: 9-13], but differ from that of Sulculus diversicolor myoglobin in 45-46 amino acids [Suzuki, T. and Takagi, T. (1992) J. Mol. Biol. 228: 698-700]. A phylogenetic tree was constructed from the amino acid sequences of 304 residues of myoglobins from N.gigantea, N.discus, N.madaka and S.diversicolor to make clear the biological positions of the abalones. The phylogenetic analyses suggested that Nordotis and Sulculus diverged at 108 million years before present (MyrBP), and that the radiation of the three Nordotis species occurred around 4-7 MyrBP.
