Overexpression of CONSTANS from Arabidopsis thaliana did not cause early flowering in the short-day tobacco plant, Nicotiana tabacum cv. Hicks Maryland Mammoth, under a long-day condition

Transgenic short-day tobacco plants, Nicotiana tabacum cv. Hicks Maryland Mammoth, were generated in which Arabidopsis thaliana CONSTANS cDNA with the Omega translational enhancer was expressed under the control of the constitutive cauliflower mosaic virus 35S promoter. In a long-day condition, 16 h light /8 h dark, the transgenic plants did not flower over 210 days of experimental period after they had been transferred to pots while control transgenic plants possessing an empty T-DNA flowered around 121 days after the transfer. This result suggests that Arabidopsis CONSTANS does not act as a positive regulator for flowering in short-day plants under long-day conditions, and supports the idea that CO orthologs in short-day plants act as a repressor for flowering under long-day conditions, similar to Hdl in rice.オメガ翻訳エンハンサー配列を有するシロイヌナズナCONSTANS cDNAをカリフラワーモザイクウイルス35Sプロモーターの制御下で恒常的に発現させた短日性タバコNicotiana tabacum cv. Hicks Maryland Mammothを作製した。16時間明期8時間暗期の長日条件で育成した場合、cDNAを組み込んでいないT-DNAを有するコントロール形質転換植物が植木鉢に移植後121日頃に開花したのに対し、シロイヌナズナCONSTANS cDNAを過剰発現した形質転換植物は移植後210日を経過しても開花しなかった。この結果は、シロイヌナズナCONSTANSが、長日条件では、短日植物の開花を正に制御する因子として働かないことを示唆しており、短日植物においてCONSTANSオーソログは長日条件下での開花を抑制する因子として働くというイネのHdlの知見から得られた考えを裏付けるものであった。
