It is the purpose of this paper to report on the data obtained in my laboratory during the past few years, the investigations having been directed toward the pharmacological studies on the adrenal cortex hormones. I described fist the chemistry of these hormones, and then the criticism of its various assay methods: survival-growth method, potassium intoxication test, liver glycogen test, anti-insulin test, swimming test, cold test, rectum temperature test, and eosinopenia method. The eosinopenia method which examines the activity of samples, determining their ability to decrease eosinophile count in circulating blood, was applicable as a most sensitive bio-assay method. In order to establish the physiology of these hormones, we tried some experiments on the relation between adrenal cortex and 1) fatigue of skeletal muscle, 2) capillary permeability, 3) permeability of frog's skin, 4) diuresis, 5) histamine and acetycholine content in blood and tissnes, 6) alloxan diabetes. Sayers & Sayers and Long suggested that epinephrine acts like other nonspecific stresses and does not have direct action upon the pituitary. But, on the contrary of their opinion, Harris et al concluded that secretion of ACTH is under neural control via the hypothalamus and the hypophyseal potal vesseis, and there is no agreement concerning the manner of this function. On the basis of the results obtained by the experiments on the phenomenon of decreasing effect of eosinophils in circulating blood, I have outlined my views concerning the interrelationship existing between the diencephalon particularly hypothalamus, pituitary and adrenal cortex, and stressed the important role played by the hypothalamus.
