Total RNA was successufully isolated from the pine wood nematodes, Bursaphelenchus xylophilus, by the SDS-phenol/CsCl method. Poly(A)+ RNA was purified from the RNA, and was shown to have template activity. Since poly(A)+ RNA is produced by eucaryotes, including the pine wood nematode, the template active poly(A)+ RNA of the pine wood nematodes can be used to clarify the origin of cellulase in the pine wilt disease.
