Poly-L-malic acid (PMA), a protease inhibitor from Penicillium cyclopium, has been shown to act not only on the acid protease of the same mold but also on the other kind of proteases. This causes the proteases to change conformationally, whereby the enzymes become more susceptible to auto-digestion which abolishes the enzyme activity. This work was undertaken to learn whether PMA might act on proteins in a similar fashion, whereby the proteins become more susceptible to proteolysis. Casein, ovalbumin, Bacillus subtitis α-amylase, and lysozyme were used as test proteins. The results indicated, contrary to expectation, that PMA did not bring about any enhancement of susceptibility to proteolysis of these proteins. It was also found that P.cyclopium acid protease・PMA complex and B.subtilis alkaline protease・PMA complex, which underwent accelerated auto-digestion as described earlier, were not more susceptible to the proteolysis by different proteases than the free enzyme.
