We reported in the previous paper that the yeast, Trichosporon X-19, produced extra cellular protein (ECP) when it was cultured in an acidic environmen. However, adoption of arginine as a nitrogen source for the medium made it possible for the yeast to produce ECP even though in a neutral environment. Having proven that high concentration glycine modified and collapsed bacterial membranes, we discussed the effect of arginine concentration on the ECP production and cell membrane construction of the yeast. To summarize our findings : arginine acted differently from glycine, that is, an arginine concentration of 0.2-0.5 % gave maximum ECP production, however in the high concentration medium only poor ECP production was observed. Observation of the yeast cells under an electron microscope (SEM and TEM)also revealed apparent abnormalities, such as protoplasmolysis, in the cells cultured in the medium containing 0.2-0.5 % arginine. In contrast, hardly any abnormalities were observed in the cells cultured in the medium containing under 0.1 % or over 1 % arginine.
