Identification of Traditional Chinese Patent Medicines(中成薬)by a Polariscope(1)Polariscopic Characteristics of Starch Grains and Calcium Oxalate Crystals

Identification of powdered crude drugs by microscopic method is not always possible as fragments of powdered crude drugs often overlap with other fragments. In this paper, we describe that polariscopy enabled easier observation of starch grains and crystals of calcium oxalate with no need of staining, because under a polariscope, starch grains showed a black cross with the center of cross in hilum, and crystals of calcium oxalate obvious brightness. By using these features, the starch grains of different origins and those in Dioscoreae Rhizoma, Cistache Herba, Astragali Radix and the crystals of calcium oxalate in Longan Arillus, Persicae Semen, Toosendan Fructus and Aurantii Fructus Immaturus were examined and accurately characterized. Polarizing microscopy was applied also to identification of ingredients of the traditional Chinese patent medicines, Liu Wei Di Huan Wan and Shen Rong Bu Xue Wan to confirm the presence of Alismatis Rhizoma, Dioscoreae Rhizoma, Moutan Cortex in the former and Longan Arillus, Ginseng Radix, Achyranthis Radix, Morindae Officinalis Radix in the latter. The characteristics of 8 starch grains and 12 crystals of calcium oxalate are listed and some polarizing color photomicrographs (×400) are also given.
