Shoot tips of Codonopsis lanceolata Trautv. (Campanulaceae) were cultured on Murashige and Skoog (MS) medium supplemented with 0.5 mg/l 6-benzylaminopurine (BAP) to give multiple shoot formation (approximately 11 shoots) without callus formation. The number of shoots formed was in inverse proportion to the BAP concentration. On the other hand, a combination of indole acetic acid (IAA) and BAP promoted the callus formation and subsequent regeneration of shoot. The number of shoots regenerated from callus culture was 4.2-6.0 at least for the following 6 generations. The stem segments formed callus when cultured in MS medium supplemented with BAP-IAA or with naphthalene acetic acid (NAA) or 2,4-dichlorophenoxy acetic acid (2,4-D). However, the number of shoots regenerated from callus was smaller than that from shoot tips. When the multiple shoot was cultured in the MS medium, with no growth regulators proper root formation occurred. After 1-month-storage of the plantlets at 5℃, they were all successfully transplanted. HPLC analysis demonstrated that the amounts of tangshenoside I and saponins (tentatively named saponin a and b) in the parent plants and in the regenerated plants were almost the same.
