Endometrial cyclic changes are well known to be regulated by ovarian steroid hormones. Estradiol stimulates endometrial proliferation and progesterone facilitates stromal decidualization. Several lines of evidence suggest that the effects of these steroidal hormones on the endometrium are mediated by the local insulin-like growth factor (IGF) system, i.e. by IGF and its binding proteins. In this study, the physiological significance of the IGF system in endometrial proliferation and decidualization was investigated. Estradiol increased the levels of IGF-I in media of cultured endometrial cells. Estradiol and progesterone promoted stromal decidualization, characterized by a specific cellular phenotype and heralded by the production of prolactin. IGF-I inhibited these effects of estradiol and progesterone. The level of IGFBP-1 in the medium increased with stromal decidualization. Progesterone increased IGFBP-1 mRNA in the endometrium. Stromal decidualization was promoted by IGFBP-1 without progesterone, but not by prolactin. We supeculate that local IGFs function in the endometrial regulation governed by ovarian steroid hormones.
