Histopathological and electron microscopic studies of the experimental gastric ulcer induced by a clamping method were performed. 1) The ulcer induced by this method had generally a tendency to heal well. The granulation tissue was observed to proliferate from 3 days on and up to 2 weeks. The granulation tissue turned into a scar tissue by 5 weeks and the ulcer was covered completely by the regenerating epithelium. Electron-microscopically, many active fibroblasts with a well developed granular endoplasmic reticulum and the accelerated formation of collagen fibrils were observed in the granulation tissue of 5 to 7 days. 2) In the animals treated with cortisone, however, the proliferation of the granulation tissue was prominently suppressed and the ulcer did not have a complete recovery until about 12 weeks. The fibroblasts in the granulation tissue of 5 to 7 days showed many ultrastructural abnormalities i. e. marked dilation of the cisternae of the granular endoplasmic reticulum, large vacuoles and lipid granules in unusually increasing numbers. The collagen fibrils were only sparsely found among the fibroblasts. Thus, the prolongation of the ulcer healing in this group was supposed to be due, at least in part, to the pathological changes of the fibroblasts with the resultant disturbance of the collagen fibril formation. 3) DOGA activated the granulation tissue proliferation and the ulcer healing was accomplished by 3 weeks.
