The long term feeding experiments were carried out to estimate chronic toxicity when rats fed with the diet added two preservatives, a new nitrofuran 2-(2-furyl)-3-(5-nitro-2-furyl) acrylamide and sorbic acid at the same time. Sixty-six Wister rats were divided into 5 groups, (1) nitrofuran 0.001% and sorbic acid 0.1%, (2) nitrofuran 0.01% and sorbic acid 1.0%, (3) nitrofuran 0.001% singly, (4) nitrofuran 0.01% singly, (5) control, and the animals were fed under the condition of 24℃ and 60% humidity for a 12-17 months period. The results obtained were following : 1. During the course of experiments, no definite signes of abnormalities for the animals were observed in each drug levels of the diet ; such as food consumption, appearance of skin and hair, state of motility and mortality. 2. From an analysis of growth curves, no significant differences were shown between the groups nitrofuran-sorbic acid combined diet and single nitrofuran diet in body weight gain. 3. Post-mortem examination in survived animals did not show macroscopically any pathological changes in liver, kidney and other main organs. Haematological tests were also shown normal limits. 4. Microscopic studies on the main organs of rats sacrificed at 12 month and 17 month were made in detail. Degenerative changes, atrophy, hydropic degeneration, pyknosis and fatty metamorphosis of hepatic cell were not demonstrated in all cases. 5. There was nothing of the neoplastic changes by the added drugs in hepatic cells of all the rats tested. 6. From the data above mentioned, it is clear that the addition of sorbic acid does not show any effect to increase of chronic toxicity of 2-(2-furyl)-3-(5-nitro-2-furyl) acrylamide.
