This paper describes the action of various drugs on the bronchial muscnlature using the technic of a modified method of Bhattacharya and Deiaunois on the isolated guinea-pig's lung perfusion preparation. As ether inhalation tends to produce bronchial hypersecretion, animal bled to death by cutting both carotid artery withont anesthesia. The Mariotte bottle and the cannula of the pulmonary artery shonld be lied on the same level. The addition of 3% dextran in the perfusion Tyrode's fluid and heparinized anto-blood in a ratio 2% was of great help to prevent oedema in the duration of the experiment. 5% CO_2+95% O_2 mixture shonld be enter into the lung through the tube, using the artificial respirometer (120/min, 150 cc/volume). Using this technic, it has been possible to work at least 3 hours or more with the lung preparation in a physiological state. Acetylcholine, histamine and serotonin produced increasing effect of the inner-pressure of the beonchus, contraction of bronchial musculature and decrease of the pressure of perfusion fluid. Adrenaline antagonise the effect of acetylcholine, histamine and serotonin on the bronchial musculature. Atropine antagonise markedly the contractile effect of bronchial musculature induced by acetylcholine and histamine but, it is not an effective antagonist against serotonin. Anergen and Diparcol antagonise the spastic effect of histamine and acetylcholine but has no influence against serotonin effect. Tn a small doses of chlqrpromazine failed to observe any influence against the effect of acetylcholine but there is an antagonistic effect to it in a more larger doses. Chlorpromazine is not .an effective antagonist against serotonin.
