The kitten test is a regular method used for detecting enterotoxin in all the world. However, the vomitings of these animals not due to enterotoxin were often observed. Therefore, the reliability of the test is rather low. The author investigated the cause of confusion by purifying the toxic substances from the culture filtrates and human volunteer tests of each extract fractions. The results obtained were as follows: 1) Vomitings and other symptoms irritating gastro-intestinal tractus were observed by the kitten tests of thirty strains, while the culture filtrates of only three strains of the group brought about these symptoms by the human volunteer tests. 2) The purification of enterotoxin was performed by the combination of four methods; salting out from the culture filtrates by the addition of ammonium sulphate, acid precipitation at pH 3.5, column-chromatography using hyflosupercel and zone-electrophoresis. 3) The toxic fractions for kitten tests were distinguished from onse for human volunteer tests, because the former fractions could not be extracted from the same band of zone-electrophoresis as the latters. 4) Utilizing the specific mobility of the enterotoxin in the electrophoresis, the chemical detection of the toxic substance was considered- to be possible.
