In the present investigation, the effect of antihistarninics in such compounds as Benaaryl, 3015 R. P, 3277 R. P has been tested on the rectus muscle of frog. The results are summarized as follows. Antihistaminics did show not only antihistaminic but also antiacetylcholinic action on the skeletal muscle contraction and the same effect was demonstrated in the earlier report of the same author's experiments on the isolated frog heart and guinea pig ileum. It appears that the site of action of acetylcholine is located mostly on the surface of the muscle tissues and that of antihistaminics not only on the surface but also in the tissues. Marked antiacetylcholinic action was observed when antihistaminics was administered in an adequate dosage. A high dosage of antihistaminics produces contractile action of the muscle and this was potentiated by acetylcholine and KCl. This effect was not influenced by curarine, atropine and tetrodotoxin and antagonizes Mg. The effect of antihistaminics on the skeletal muscle was closely related with the effect of KCl, but it is not quite the same because the pretreatment of antihistaminics depresses the contractile action of KCl.
