In order to develop a DNA extraction method suitable for the practice in molecular biology for our students, who intend to be primary school teachers, we investigated the condition of DNA extraction from chicken liver without using harmful reagents and a centrifuge. Ten % (w/v) liver homogenate in Tris-buffered saline was heated in boiling water for 3 min in the presence of 1.5M NaCl and 1.0% (w/v) sodium dodecyl sulfate, followed by the immediate chill on ice. The suspension was filtered through 3 layers of gauze, and then 2.5 vol. of cold ethanol was overlaid onto the filtrate. The fibrous DNA appeared at the interface was wound on a glass bar. The yield of DNA was 1-1.5 mg/g of liver. In a pilot practice, the students successfully extracted significant amounts of DNA from 2 g of liver, and they realized the DNA to be one of the components in biomaterials.
