Effects of traditional Japanese (Kampo) medicines (orengedokuto, goreisan and shichimotsukokato) on the onset of stroke and expression patterns of plasma proteins in spontaneously hypertensive stroke-prone rats

脳卒中易発症自然発症高血圧ラット(SHRSP)を用いて,脳卒中発症と生存率ならびに脳卒中発症前後の血漿蛋白の発現に及ぼす漢方薬の効果を検討した。方法は7週齢の雄のSHRSP32匹を対照群(蒸留水を自由飲水),黄連解毒湯群(0.3%黄連解毒湯エキス水溶液を自由飲水),五苓散群(0.3%五苓散エキス水溶液を自由飲水),七物降下湯群(0.3%七物降下湯エキス水溶液を自由飲水)の4群に分け,20週齢まで飼育した。12と14週齢時に血圧を測定し,1週間毎に体重測定と尾静脈からの採血を実施した。血液検体は血漿プロテオーム解析を施行した。結果は,脳卒中の発症後に認められる体重減少に関して,対照群は14週齢以後体重減少を認めたが,黄連解毒湯群,五苓散群,七物降下湯群は,20週齢の時点で体重減少を認めなかった。収縮期血圧は14週齢では4群間で差を認めなかった。また,生存率は,黄連解毒湯群,五苓散群,七物降下湯群で対照群に比べて有意な改善を認めた。血漿蛋白発現の解析ではm/z(質量/荷電)3,000-30,000の間で,対照群で脳卒中の発症に伴って変動するピークを15個認めた。特に脳卒中発症後にm/z9,330,9,480,9,700のピークは著しく減少した。これらのピークはWestern blot法によりハプトグロビンであることをすでに明らかにしている。一方,これらのピークは漢方薬投与群で減少が抑制された。以上のことから,黄連解毒湯,五苓散,七物降下湯は,血圧への関与以外の作用機序で,SHRSPの脳卒中発症を抑制し,脳卒中発症に関連する蛋白成分に影響を及ぼしている可能性が示唆された。
