Hepatitis B virus (HBV) genotypes induce liver disease of distinct severity, but the fundamental virological differences are not well defined. In our recent studies using Huh7 cells and chimeric mouse harboring human hepatocytes, we demonstrated the particular characteristics of each genotype although it was general that virological properties of HBV genotypes were almost similar to each other. We have cloned 1.2-fold the HBV genome of different genotypes/subgenotypes from serum samples. HBV DNA levels in cell lysates were the highest for HBV/C and B and the lowest for HBV/A and D. HBe antigen and HB core protein were excreted in a trend similar to that of HBV DNA with similar differences. Secretion of HB surface antigen, however, was the most abundant HBV/A followed by B, C and remotely by D, which was consistent with mRNA levels. Chimeric mouse with the liver replaced for human hepatocytes were inoculated with virion passed in mouse. HBV DNA levels in their sera were higher for HBV/C than A by 2 logs during 4-7 weeks after inoculation. Virological differences among HBV genotypes were demonstrated both in vitro and in vivo. These characteristics might influence HBV infection in clinical and epidemiological setting.
