Karyotype analysis of a Japanese cucumber cultivar by fluorescence in situ hybridization

Well-spread prometaphase chromosome preparations of a Japanese cucumber cultivar 'Jibai' were observed and karyotype analysis was carried out on the basis of morphology, density patterns stained with Giemsa and DAPI, and rDNA loci. Density profiles of the chromosomes stained with Giemsa and DAPI were analyzed by using software CHIAS III (Chromosome Image Analyzing System III) and ideograms were constructed according to the density profiles. The 45S and the 5S rDNA FISH signals were detected on three and two pairs of chromosomes, respectively, and were located on the ideograms. From these results, seven pairs of chromosomes were discriminated with their morphology and unique density patterns, and were designated as chromosomes 1 to 7 in order of their total lengths.
