Microcystins, the cyclic heptapeptides, are produced by some cyanobacteria belonging to genera Microcystis, Anabaena, Nostoc, and Oscillatria. They show potent acute hepatotoxicity and tumor-promoting activity to livestock and human through inhibition of protein phosphatases (PP) 1 and 2A. Microcystins are stable against various physicochemical factors because of its structural characteristic features. Therefore, a regulation method has been required for microcystins under natural conditions. We are developing a control method for microcystins using co-existing microorganisms in the same ecosystem and isolated a bacterium that can degrade 1, designated B-9 strain, belonging to a new genus related to Sphingomonas, from Lake Tsukui. We have confirmed that B-9 strain degrades 1 with intracellular enzymes. As a result of the observation of microcystin degradation process using B-9 strain by ESI-LC/MS, several degradation products were concurrently detected with the degradation of 1 and 4 was finally accumulated. As reported by Bourne et al., it was confirmed that 2 and 3 were detected as the intermediates by the use of protease inhibitors, PMSF and by EDTA, respectively. To reveal the substrate specificity of microcystin-degrading enzyme of B-9 strain, we observed degradation behavior of microcystin related compounds. It was found that 5, 7, and 11 were quickly degraded as well as 1, but 6, 8, 9, and 10 were not completely degraded after 72 hr. These results show that the degradation of microcystins and related compounds by B-9 strain depends on the structures of Mdha and Adda portions, and the presence or absence of L-Arg.
