Soybean seeds contain many kinds of saponins and some of them are considered to have defence effect on HIV (human immunodeficiency virus). Generally. saponins are classified to three groups (group A, B, and E) according to their chemical structures. "Group A" saponins have two suger chains which link to C-3 and C-22 aglycone structures. "Group B" and "Group E" have a single chain which linkes to C-3. It is of great importance to clarify the structures of these soybean saponins. In this work, we studied the structures of "Group B" saponins by using by Liquid Chromatograhy/Mass Spectrometry (LC/MS) technique. We isolated five kinds of saponins named soyasaponins a g, αg, βa, γg, and γa, according to elution order from HPLC. Then the structures were characterized as 2,3-dihydro-2,5-dihydro-2,5-dibydroxy-6-methyl-4h-pyran-4-one (DDMP) attaching through an acetal linkage to the C-22 hydroxyl of the aglycones of "Group B" saponins. We also found a possible mechanism in the formation of the suger sequence by using the same LC/MS technique.
