67 Streptothricin抗生物質の生合成に関する研究

The study was made of the biosynthesis of streptothricin antibiotics. The results are as follows: 1) D-glucosamine 1-^<14>C was incoporated into D-gulosamine moiety of racemomycin-A which was produced by Streptomyces lavendulae ISP-5069. 2) Streptolidine moieties were derived from L-arginine-U-^<14>C. Furthermore, it was proved that arginine labeled with ^<14>C in C-1, C-5 and aminido group were incorporated well into streptolidine moieties. The incorporation of arginine into streptolidine of streptothricins by the fermentative production of four Streptomyces strains was then investigated. As result, arginine was also found to be incorporated appreciably into streptolidine by other Streptomyces except for Streptomyces lavendulae ISP-5069. 3) In a previous study of biosynthetic pathway leading to the formation of racemomycin-A, we obseved that ^<14>C from arginine was not located in the streptolidine moiety in this antibiotc, whereas the ^<14>C from (1-^<14>C) acetate was. However, we did not determine the distribution of labeled atoms in the metabolite. The availability of CMR spectroscopy promoted us to use ^<13>C-labeled acetate for this purpose. As result, it is evident that (1-^<13>C) acetate was incorporated into the carbons C-1 and C-5, and that (2-^<13>C) acetate was incorporated into C-2 and C-4 respectively.
