Neurite promotive effect of nipradilol on adult rat retinal ganglion cells

Objective : To investigate the effects of nipradilol on neurite regeneration in adult rat retina. Methods : Retinas, dissected from adult rats were cultured in a collagen gel with serum free media. The final concentrations of 10^<-10> to 10^<-5>M nipradilol were added to each medium. The capabilities of neurite regeneration were evaluated by counting the outgrowing neurites from the retinal explants. Results : Numbers of regenerating neurites in each concentration at 9 days were 26.7(10^<-10>M), 28.3(10^<-9>M), 37.6(10^<-8>M), 59.1(10^<-7>M), 47.3(10^<-6>M) and 26.0(10^<-5>M) per explant, respectively. The application of 10^<-7>M nipradilol significantly increase the neurite numbers approximately two-fold compared to that of the control (culture without nipradilol) at 9 days in culture. Conclusion : Nipradilol has a neurite regenerative effect on adult rat retinal ganglion cells.
