Temporal variations in the pharmacokinetics and metabolism of theophylline were investigated in seven healthy volunteers following single oral dosing of an immediate-release theophyiline tablet formulation at 9:00 a.m. and 9:00 p.m. Theophylline concentration in the plasma and theophylline and its metabolites excreted in the urine were measured by a high-performance liquid chromatographic (HPLC) method. The pharmacokinetic and bioavailability parameters obtained in this study showed no difference between morning and evening dosing. Cumulative urinary excretion and urinary recovery of theophylline and its metabolites also showed no difference between morning and evening dosing. The present study shows that there are no temporal variations in the pharmacokinetics and metabolism of theophylline.
