Glycosylated hemoglobin (Hb) formed by glycosylation between glucose and Hb. In present study, I investigated glycosylated hormone using 3, 5, 3'-L-triiodothyronine (T_3). The measurement and separation of T_3 and glycosylated T_3 (G-T_3) were performed by high performance liquid chromatography (HPLC) on a μBondapak C18 column using paired-ion chromatography (PIC) B6 in acetonitrile solution (acetonitrile and water in the ratio 40 : 60) as a solvent. The biological activities of T_3 and G-T_3 were assayed using the metamorphosis of tadpoles (Rana nigromaculata). The results for the above experimental studies were obtained as follows : 1) In the reaction between T_3 and glucose in vitro, the production rate of G-T_3 increased in proportion to reaction time and glucose concentrations. 2) In the measurement of T_3 and G-T_3 using RIA, no significant difference was observed between them. 3) It was able to be proved using thin layer chromatography that G-T_3 is produced by incubation experiment of T_3 and glucose. 4) The biological activity of G-T_3 was less than about 1/10 of that of T_3 at their concentration of 3×10^<-6>mM. These findings suggest that Ta is nonenzymatically glycosylated and its biological activity is decreased. As these experimental results, it appears that special attention should be paid to be heterogeneity of various hormones in diabetic state with hyperglycemia.
