To investigate the role of intracellular Ca^<2+> in the mechanism of cellular proliferation of vascular smooth muscle cells (VSMC), the effects of Ca^<2+>-antagonists and calmodulin (CaM) inhibitors on DNA synthesis were studied in VSMC from rat aorta. Among several serum-derived growth factors, both epidermal growth factor (EGF) and platelet-derived growth factor (PDGF) significantly stimulated DNA synthesis by VSMC. Ca^<2+>-antagonists (verapamil, diltiazem and nicardipine) significantly inhibited DNA synthesis stimulated by EGF, PDGF and fetal bovine serum in a dosedependent manner (10^<-6>〜10^<-5>M). CaM inhibitors (trifluoperazin and W-7: 10^<-6>〜10^<-4>M) were also effective in inhibiting DNA synthesis, whereas W-5 was minimally effective. These data suggest that Ca^<2+>-CaM system plays an important role in the regulation of DNA synthesis by VSMC.
