YRFB (H-Tyr-D-Arg-Phe-βAla-NH_2) is a short analogue of dermorphin with high activity at the μ opioid receptor. In the present study, two YRFB analogues containing 2', 6'-dimethylphenylalanine (Dmp) in position 1 or 3 were examined for their antinociceptive activities by the formalin test in mice after subcutaneous injection. Both [Dmp^1] YRFB and [Dmp^3] YRFB produced dose-dependent antinociceptive activities in the formalin test. [Dmp^3] YRFB exhibited activities about 40-fold and 70-fold more potent than morphine at the first and second phases, respectively. [Dmp^1] YRFB exhibited twice and 5-fold as potent as morphine at the first and second phases, respectively. These results suggest that the Dmp-containing analogues are useful analgesics for the in vivo studies of the opioid system.
