Cytokine Dependency of Granzyme B mRNA Expression in Natural Killer Leukemic Cell Line, YT-u

Cytokine dependency of granzyme B (grB) mRNA expression was investigated using RT-PCR. First, expression of grB mRNA in T and B cells, a natural killer leukemic cell line (YT-u) and megakaryocytic cells were examined. GrB mRNA expression was detected in CD8^+ T cells and YT-u, but not in other hematopoietic cells. GrB mRNA expression in YT-u was augmented by the addition of 5-400 U/ml IL-2 and 2-20 U/ml IL-12 in a concentration-dependent manner. Intense augmentation was observed 12 hours after the addition of either IL-2 or IL-12, and the intensity was quantitatively determined with competitive RT-PCR. The number of copies of grB mRNA was 1 × 10^6 in non-treated YT-u, 5 × 10^6 copies after the addition of 50 U/ml of IL-2 or of 2 U/ml of IL-12 and 1 × 10^7 copies after the addition of a combination of IL-2 and IL-12. Expression of grB mRNA in normal NK cells has been reported to increase synergistically after the addition of IL-2 and IL-12. It therefore seems possible that YT-u may have a different intracellular signal transduction system for cytokine-induced regulation of grB mRNA from that of normal NK cells.
