Autoimmune-Neutropenia : Characterization of Antineutrophil Autoantibody

• We detected anti-neutrophil autoantibody in a serum from a chronic neutropenic adult patient and attempted characterization of target antigens by serological and immunochemical methods. The serum was found to react to neutrophils from 13 out of 17 healthy donors. This finding suggested the high incidence of the target antigen expression on human neutrophils. Serological tests for reactivity to neutrophil alloantigen (NA 1, NA 2, NB 1, NB 2, 5 b and Mart) showed no specificity to these known alloantigens. Through immunoblotting analysis, two bands of molecular weight 80 kDa and 50 kD were detected by the patient's serum.
