Improvement of Lectin Binding by Microwave Irradiation. : Histochemistry of Amaranthus caudatus, Arachis hypogaea, Vicia villosa and Limax flavus

We previously proved that the positive rate of Griffonia simplicifolia agglutinin-1 binding sites was increased by microwave irradiation (MWI). In this study, we applied MWI to improve the unstable staining pattern of four kinds of lectin. The positive rate on the apical surface was checked. By the conventional method, Amaranthus caudatus agglutinin (ACA) binding sites were positive in 1/6 carcinomas and Arachis hypogaea agglutinin (PNA) binding sites, in 3/6 carcinomas. In adenoma and non-neoplastic mucosa, neither ACA nor PNA binding sites were recognized. After MWI, ACA binding sites improved positively up to 3/6 in carcinomas, 4/6 in adenomas and 1/6 in non-neoplastic mucosa. PNA binding sites also improved positively up to 6/6 in carcinomas and 2/6 in adenomas, but remained negative in non-neoplastic mucosa. Vicia villosa agglutinin (VVA) binding sites were detected in 1/6 carcinomas by the conventional method, and increased up to 3/6 in carcinomas after MWI. VVA binding sites were recognized in neither adenoma nor non-neoplastic mucosa. By the conventional method, Umax flavus agglutinin (LFA) binding sites were positive in 1/6 carcinomas and non-neoplastic mucosa, respectively, but were negative in adenoma. After MWI, the positive rate increased up to 2/6 in carcinomas, 1/6 in adenomas and 1/6 in non-neoplastic mucosa. MWI was effective in improving the positive rate of binding sites of these lectins.
