The purification of aminopeptidase I was carried out on the crude extract which was obtained from seedlings of 6 days' germination by ammonium sulfate precipitation (30-70%), column chromatographies on DEAE-Sepharose and Sephadex G-150. The molecular weight of this enzyme was estimated to be about 55,000 by the gel filtration method. The optimal pH and temperature of the enzyme were at 7.0 and 50℃, respectively. Even though the enzyme solution was let stand at pH 5-9 for 2 hour at room temperature, the enzyme activity was comfirmed to be the same, and below 40℃, the solution was stable against the heat treatment for 10 min. The enzyme activity was inhibited by Ag^+, Hg^+ and Cu^<++>, but noinhibited by several reagent. As for the substrate specificity of this enzyme, leucyl-jS-naphthyl-amide was the most suitable substrate of those tested.
