This goal of this study was to characterize the role of endothelium-derived relaxing factor (EDRF)/nitric oxide (NO) in the attenuation of the hypoxia (3% O_2)- and angiotensin II (All, 0.2μg)-induced pulmonary vasoconstriction after exercise training. Further, this study investigated whether the flow (12 to 32.5mL/min)-induced endothelium-mediated dilation of pulmonary vasculature is altered after exercise training in isolated rat lungs perfused with a physiological saline solution. Twenty-four male Sprague-Dawley rats were assigned to one of two groups : a control group and an exercise-trained (ET) group. ET rats were trained on a treadmill at a running speed of 20-30m/min for 20-40 min/day, 5 days a week for a total of 8 weeks. Experiments demonstrated that exercise training resulted in a reduction in hypoxic pulmonary vasoconstriction (HPV). Moreover, administration of N^G-nitro-L-arginine methyl ester (L-NAME, 1.5×10^<-8>M) resulted in increased HPV in ET rats when compared with control rats. Exercise training also attenuated All-induced pulmonary vasoconstriction, and L-NAME administrated resulted in an increase in All-induced pulmonary vasoconstriction in ET rats when compared with control rats. Further, following L-NAME administration, pulmonary arterial pressure was significantly higher in ET rats than in control rats, despite a corresponding increase in the perfusate flow. These data suggest that exercise training promotes endothelium-dependent vasodilation via increases in NO in the context of pulmonary vasoconstriction and increased perfusate flow.
